Course objectives

  The main objectives of the module is:

  • to provide the methodologies to conjugate imaging probes to immune-based systems
  • to provide a comprehensive understanding of imaging probes based on antibodies, antibody fragments or derivatives, and antibody mimetics toi be used in in vivo imaging technologies
  • to familiarize students with the different types of immune-based imaging agents and their applications in disease detection and monitoring.
  • to examine the advancements and challenges in the development and translation of immune-based imaging agents from preclinical to clinical settings.

Results of learning outcomes

Students will acquire an integrated comprehension of the design and use of immune-based imaging probes for in vivo molecular imaging procedures  for diagnostic and theranostic purposes.

Introduction: use of immuno-based agents in diagnosis and theranosis

Bioconjugation: low-specific methods (amino groups)
 
Bioconjugation: site-specific methods (cysteine – glycans – peptide tags – unnatural and noncanonical aminoacids)
 
Use of antibodies in molecular imaging (target: HER2 – EGFR – FR -  PSMA – CD20 – PD1/PDL1)
 
Use of Antibody-Drug Conjugates in molecular imaging
 
Antibody-based structures in molecular imaging (Ab fragments – Minibody – Diabody - Nanobody)
 
Antibody mimetics in molecular imaging (DARPins – Monobodies – Anticalins – Affibodies – Affimers)
 
Examples of immuno-based imaging by Optical/MRI/US technologies

The lessons will be accompanied by material uploaded online on the Moodle platform. The students are invited to interact with the instructor by posing and answering questions

Learning assessment methods

Learning will be assessed by a written examination in which students will be asked questions about the main topics of the course. The use of correct terminology and clarity of the exposition will be also evaluated.
